Systemic kappa opioid receptor antagonism accelerates reinforcement learning via augmentation of novelty processing in male mice

Zahra Z. Farahbakhsh, Keaton Song, Hannah E. Branthwaite, Kirsty R. Erickson, Snigdha Mukerjee, Suzanne O. Nolan & Cody A. SicilianoÂ

doi : 10.1038/s41386-023-01547-x

Selective inhibition of kappa opioid receptors (KORs) is highly anticipated as a pharmacotherapeutic intervention for substance use disorders and depression. The accepted explanation for KOR antagonist-induced amelioration of aberrant behaviors posits that KORs globally function as a negative valence system; antagonism thereby blunts the behavioral influence of negative internal states such as anhedonia and negative affect.

Does cannabidiol make cannabis safer? A randomised, double-blind, cross-over trial of cannabis with four different CBD:THC ratios

Amir Englund, Dominic Oliver, Edward Chesney, Lucy Chester, Jack Wilson, Simina Sovi, Andrea De Micheli, John Hodsoll, Paolo Fusar-Poli, John Strang, Robin M. Murray, Tom P. Freeman & Philip McGuireÂ

doi : 10.1038/s41386-022-01478-z

As countries adopt more permissive cannabis policies, it is increasingly important to identify strategies that can reduce the harmful effects of cannabis use. This study aimed to determine if increasing the CBD content of cannabis can reduce its harmful effects.

doi : 10.1038/s41386-022-01412-3

Cyclin-dependent kinase-like 5 (CDKL5) deficiency disorder (CDD) is a devastating rare neurodevelopmental disease without a cure, caused by mutations of the serine/threonine kinase CDKL5 highly expressed in the forebrain.

Altered neuronal activity in the ventromedial prefrontal cortex drives nicotine intake escalation

Myriam Abarkan, Giulia R. Fois, Caroline Vouillac-Mendoza, Serge H. Ahmed & Karine GuillemÂ

doi : 10.1038/s41386-022-01428-9

Nicotine addiction develops after prolonged drug use and escalation of drug intake. However, because of difficulties in demonstrating escalation of nicotine use in rats, its underlying neuroadaptations still remain poorly understood.

Anandamide and 2-arachidonoylglycerol differentially modulate autistic-like traits in a genetic model of autism based on FMR1 deletion in rats

Sara Schiavi, Antonia Manduca, Emilia Carbone, Valeria Buzzelli, Alessandro Rava, Alessandro Feo, Fabrizio Ascone, Maria Morena, Patrizia Campolongo, Matthew N. Hill & Viviana TrezzaÂ

doi : 10.1038/s41386-022-01454-7

Autism spectrum disorder (ASD) has a multifactorial etiology. Major efforts are underway to understand the neurobiological bases of ASD and to develop efficacious treatment strategies. Recently, the use of cannabinoid compounds in children with neurodevelopmental disorders including ASD has received increasing attention.

Rostromedial tegmental nucleus nociceptin/orphanin FQ (N/OFQ) signaling regulates anxiety- and depression-like behaviors in alcohol withdrawn rats

Wenfu Li, Zhiheng Ren, Ying Tang, Yixin Fu, Shizhu Sun, Ruxuan Ding, Jiawei Hou, Yunlin Mai, Bo Zhan, Yingxin Zhu, Wanhong Zuo, Jiang-Hong Ye & Rao FuÂ

doi : 10.1038/s41386-022-01482-3

Recent studies indicate that stimulation of the rostromedial tegmental nucleus (RMTg) can drive a negative affective state and that nociceptin/orphanin FQ (N/OFQ) may play a role in affective disorders and drug addiction. The N/OFQ precursor prepronociceptin encoding genes Pnoc are situated in RMTg neurons.

Male-selective effects of oxytocin agonism on alcohol intake: behavioral assessment in socially housed prairie voles and involvement of RAGE

Sheena Potretzke, Yangmiao Zhang, Ju Li, Kristopher M. Fecteau, David W. Erikson, Marcel Hibert & Andrey E. RyabininÂ

doi : 10.1038/s41386-022-01490-3

Targeting the oxytocin (OXT) peptide system has emerged as a promising new approach for the treatment of alcohol use disorder (AUD). However, further advancements in this development depend on properly modeling various complex social aspects of AUD and its treatment.

Nicotine dependence and insula subregions: functional connectivity and cue-induced activation

Dara G. Ghahremani, Jean-Baptiste F. Pochon, Maylen Perez Diaz, Rachel F. Tyndale, Andy C. Dean & Edythe D. LondonÂ

doi : 10.1038/s41386-023-01528-0

Nicotine dependence is a major predictor of relapse in people with Tobacco Use Disorder (TUD). Accordingly, therapies that reduce nicotine dependence may promote sustained abstinence from smoking. The insular cortex has been identified as a promising target in brain-based therapies for TUD, and has three major sub-regions (ventral anterior, dorsal anterior, and posterior) that serve distinct functional networks.

Oxytocin effects on amygdala reactivity to angry faces in males and females with antisocial personality disorder

Haang Jeung-Maarse, Mike M. Schmitgen, Ruth Schmitt, Katja Bertsch & Sabine C. HerpertzÂ

doi : 10.1038/s41386-023-01549-9

The amygdala is a key region in current neurocircuitry models of reactive aggression as it is crucially involved in detecting social threat and provocation. An increased amygdala reactivity to angry faces has been reported in aggression-prone individuals and the neuropeptide oxytocin (OT) could dampen anger-related amygdala reactivity in a number of mental disorders.
