Pathogenesis of acral (localized) peeling skin syndrome

(A) TGM5 is normally expressed in the spinous and granular layers of the epidermis, where it cross-links a variety of structural proteins to form the cornified cell envelope.
(B) Biallelic mutations in TGM5, which abolish its activity, result in separation between the granular and corneal layers of the epidermis.
